Missiroli's academic background consisted of graduating from the prestigious Accademia d'Arte Drammatica in Rome, where he honed his skills in direction.

The 1950s saw Missiroli working as an assistant director at the renowned Piccolo Teatro in Milan, under the guidance of the illustrious Giorgio Strehler. In the world of cinema, he began his career as an assistant director to Valerio Zurlini.

In 1963, Missiroli made his directorial debut with the film La belle di Lodi, an adaptation of Alberto Arbasino's novel, starring the talented Stefania Sandrelli.
